Description

SPDB-DM4 is a type of antibody-drug conjugate (ADC) consisting of a cytotoxic drug payload, DM4 (derivative of maytansine), linked to an antibody via a linker molecule known as SPDB (N-succinimidyl 4-(2-pyridyldithio)butanoate). SPDB is a linker molecule that facilitates the attachment of DM4 to the antibody. It contains a reactive group (succinimidyl ester) that forms a stable covalent bond with primary amines on the antibody, and a pyridyldithio group that enables conjugation to the DM4 payload. DM4 is a potent cytotoxic agent derived from the natural product maytansine. It disrupts microtubule assembly, leading to cell cycle arrest and apoptosis, making it an effective anti-cancer agent. SPDB-DM4 offers the potential for targeted cancer therapy, allowing for the selective delivery of cytotoxic agents to tumor cells while sparing healthy tissues, thereby minimizing systemic toxicity and improving therapeutic efficacy.
