Azide-PEG8-Val-Cit-PABC-SN-38 is a cleavable ADC linker with the SN-38 payload. Azide can be used as a reactive handle for bioconjugation reactions. It allows for the attachment of other molecules, such as polyethylene glycol (PEG) or linkers, to the ADC. Val-Cit is a linker molecule that connects the antibody to the cytotoxic drug payload. It plays a crucial role in stabilizing the ADC and controlling the release of the cytotoxic drug once the ADC is internalized by the cancer cell. PABC (p-aminobenzyl carbamate) is another type of linker molecule used in ADCs. It connects the Val-Cit linker to the cytotoxic drug payload and helps in stabilizing the ADC. PABC linkers are designed to facilitate the release of the cytotoxic drug within the cancer cell. SN-38 is the cytotoxic drug payload, which is a derivative of irinotecan. It inhibits topoisomerase I, leading to DNA damage and cell death in cancer cells.
