Description

TCO-carbohydrate refer to carbohydrates (sugars or polysaccharides) that have been chemically modified with a trans-cyclooctene (TCO) moiety. TCO is a strained alkene that participates in highly selective and rapid bio-orthogonal reactions, especially in inverse electron-demand Diels–Alder (IEDDA) reactions with tetrazines. TCO-carbohydrates can be used for highly selective and efficient bio-orthogonal labeling of glycan structures in living cells or tissues. It can be incorporated into cells through metabolic glycan labeling or chemical conjugation. In addition, TCO-carbohydrates play a vital role in imaging glycans, which are important in cell signaling, immune responses, and disease progression (e.g., cancer). By incorporating TCO-labeled sugars into the glycans of live cells, researchers can visualize these complex structures using tetrazine-linked fluorescent probes.
