Ac4ManNAz (N-azidoacetylatedmannosamine-tetraacylated) is a chemical derivative of N-acetyl-D-mannosamine (ManNAc), a monosaccharide that serves as a precursor in the biosynthesis of sialic acid in cells. The peracetylation (Ac4) of the hydroxyl groups increases the molecule’s hydrophobicity, facilitating its transport across cellular membranes. Once inside the cell, Ac4ManNAz is deacetylated by cellular esterases, releasing ManNAz, which is incorporated into glycoproteins and glycolipids, where the azide group can be selectively tagged using bioorthogonal click chemistry. Ac4ManNAz is commonly used in metabolic glycoengineering to label glycans on the cell surface. Once inside the cell, Ac4ManNAz is converted into ManNAz and subsequently incorporated into glycoproteins and glycolipids during glycan biosynthesis. The azide group on the glycans can then be tagged with fluorescent probes or other bioorthogonal functional groups using click chemistry, allowing for the visualization and tracking of glycan structures in live cells.
