Biotin is a small molecule that binds with very high affinity to streptavidin and avidin, with a dissociation constant (Kd) in the femtomolar range. This strong and specific interaction is widely used in biochemical applications. PEG enhances the solubility and flexibility of the reagent, reducing steric hindrance and improving the efficiency of bioconjugation reactions. The amido linkage provides a stable and flexible connection between the biotin moiety and the maleimide group. The maleimide group reacts specifically with thiol groups (-SH) on cysteine residues in proteins, forming a stable thioether bond.
