Biotinyl Tyramide ((3aS,4S,6aR)-Hexahydro-N-[2-(4-hydroxyphenyl)ethyl]-2-oxo-1H-thieno[3,4-d]imidazole-4-pentanamide, BP, Biotin-Phenol) is a powerful tool in molecular biology and histology, enabling sensitive detection and localization of antigens and nucleic acids in tissues and cells through signal amplification techniques like TSA. Tyramide is a small molecule that can be enzymatically activated to form a reactive intermediate (tyrosyl radical) in the presence of hydrogen peroxide (H₂O₂). This intermediate can covalently bind to nearby tyrosine residues on proteins or other molecules in a process called tyramide signal amplification (TSA). Biotinyl tyramide is commonly used in IHC to amplify the signal of target antigens. In this technique, primary antibodies bind to specific antigens in tissue sections. After washing away unbound antibodies, a secondary antibody conjugated to horseradish peroxidase (HRP) is applied. HRP catalyzes the oxidation of biotinyl tyramide in the presence of H₂O₂, resulting in the deposition of biotinylated tyramide at the antigen site.
