Boc-Gly-Gly-Gly-PEG-methyltetrazine is a compound composed of multiple functional groups, each with its own significance in chemical synthesis and bioconjugation. Boc protects the N-terminal amino group of the glycine residue from unwanted reactions during peptide synthesis. Glycine is the simplest amino acid with a hydrogen atom as its side chain. It is often used as a spacer or linker in peptide synthesis due to its small size and minimal interference with protein structure. Tetrazines are highly reactive towards strained alkenes, particularly trans-cyclooctenes (TCOs), through the inverse-electron-demand Diels-Alder (IEDDA) reaction. This reaction is bioorthogonal, meaning it can occur selectively in biological environments without interfering with native biological processes.
