Description

D-Desthiobiotin or 5-Methyl-2-oxo-4-imidazolidinehexanoic acid is a structural analog of biotin that lacks the sulfur atom in the imidazole ring. D-Desthiobiotin is used in affinity chromatography columns to purify biotin-binding proteins such as avidin, streptavidin, and biotin-binding antibodies. The lower affinity of desthiobiotin allows reversible binding, enabling elution under milder conditions compared to biotin. In competitive binding assays, D-Desthiobiotin competes with biotin for binding to streptavidin or avidin. This property is utilized to assess the binding specificity and affinity of biotin analogs or inhibitors.
