Description

DM1-SMCC plays a crucial role in the development of ADCs for targeted cancer therapy, enabling the selective delivery of cytotoxic payloads to cancer cells while sparing normal tissues. DM1, also known as mertansine, is a potent cytotoxic compound derived from maytansine, a natural product originally isolated from the plant Maytenus serrata. DM1 exerts its cytotoxic effects by binding to tubulin and inhibiting microtubule assembly, leading to disruption of the mitotic spindle apparatus and ultimately, apoptosis (programmed cell death) in cancer cells. SMCC is a heterobifunctional crosslinking agent commonly used in bioconjugation chemistry. It contains two reactive groups: a maleimide group and an NHS (N-hydroxysuccinimide) ester group. The maleimide group reacts specifically with thiol (-SH) groups on proteins, forming stable thioether bonds, while the NHS ester group reacts with primary amines (-NH2) on proteins, forming stable amide bonds.
