dNTP-PEG3-4TCO (2′-Deoxyribonucleoside triphosphate-PEG3-4trans-cyclooctene) refers to a 2′-deoxyribonucleoside triphosphate (dNTP) modified with a polyethylene glycol (PEG) linker and a trans-cyclooctene (TCO) group. The PEG3 or PEG4 linker typically consists of 3 or 4 ethylene glycol units (-CH2CH2O-) that separate the nucleotide from the TCO moiety. This design provides flexibility and reduces steric hindrance, allowing the TCO group to participate more efficiently in bio-orthogonal reactions, especially the inverse electron-demand Diels-Alder (IEDDA) reaction with tetrazines. This combination has been widely used in DNA labeling, imaging, nanotechnology, and drug delivery.
