Description

DOTA-NHS-ester (1,4,7,10-Tetraazacyclododecane-1,4,7,10-tetraacetic acid mono-N-hydroxysuccinimide ester) combines the DOTA chelating agent with the NHS-ester functional group, enabling the conjugation of DOTA to primary amine-containing molecules, such as peptides, proteins, or antibodies, through amide bond formation. DOTA stands for 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid, which is a macrocyclic chelating agent known for its high stability and selectivity for binding various metal ions, particularly trivalent metal ions such as gadolinium (Gd3+), lutetium (Lu3+), and yttrium (Y3+). NHS-ester (N-hydroxysuccinimide ester) is a common functional group used for the selective modification of primary amines on biomolecules, such as lysine residues or the N-terminus of proteins. When reacted with an amine group, NHS-ester forms a stable amide bond.
