Mal-PEG1-Val-Cit-PAB-OH is a compound used in the synthesis of antibody-drug conjugates (ADCs), which are a class of targeted cancer therapies. Maleimide is a reactive functional group that selectively reacts with thiol (-SH) groups, forming stable thioether bonds. Maleimide is commonly used in bioconjugation chemistry to attach molecules to cysteine residues on proteins or peptides. Valine is one of the 20 standard amino acids found in proteins. It is used here as a linker between the PEG spacer and the cytotoxic payload to provide flexibility and minimize potential steric hindrance. Citrulline is a non-standard amino acid that can serve as a linker in ADCs. It is often chosen for its stability in circulation and its ability to be cleaved by lysosomal enzymes, releasing the cytotoxic payload inside cancer cells. PAB is a moiety commonly used to attach cytotoxic payloads to antibodies or other targeting molecules in ADCs. The PAB linker can be cleaved by lysosomal enzymes, releasing the cytotoxic payload specifically within cancer cells upon internalization of the ADC.
