N3-kethoxal (Azide-kethoxal / 3-(2-azidoethoxy)-1,1-dihydroxybutan-2-one) is a derivative of kethoxal (2-ethoxy-1,2-dihydroxyethylidene) with an azide group attached to its structure. Kethoxal is known for its ability to modify nucleic acids, specifically guanine bases in RNA, by forming a reversible adduct. N3-Kethoxal retains the ability to react with guanine bases in RNA, forming a reversible cyclic adduct. This modification is useful for probing RNA structure and dynamics, as it allows for selective labeling and can be reversed under mild conditions to restore the native RNA structure. By modifying specific guanine residues, N3-kethoxal can be used to study RNA-protein interactions. The azide group allows for further functionalization or tagging, providing a means to isolate or identify RNA-binding proteins or to study the structure and function of RNA-protein complexes.
